Abstract


The protocols evaluated included the CTAB method, Sodium Dodecyl Sulfate (SDS Method) and Sarkosyl Method without using liquid nitrogen. All three protocols evaluated effectively to isolate the DNA from each of the four genotypes of finger millet. The quality and quantity of the DNA extracted was compared through agarose gel electrophoresis as well as using UVspectrophotometer. It was found that CTAB and SDS both the methods are giving results at par with superiority of SDS over CTAB in the amplification of PCR products.
